Effects of sucrose and irradiance on germination and early gametophyte growth of the endangered tree fern Dicksonia sellowiana Hook (Dicksoniaceae) Acta Botanica

It is an endangered tree fern that grows in mixed umbrophylus forests in the tropics. Sterilized spores were germinated at 25 ± 2ºC under a 16-hour photoperiod, in Dyer and MS medium supplemented with 0 to 5% sucrose. The germination was lower with the addition of sucrose. Dry mass was higher in 30-day-old gametophytes cultivated in Dyer medium with the addition of 3 to 5% of sucrose. The dry mass decreased in 30-day-old gametophytes cultivated in Dyer and MS media without sucrose and in MS medium with the addition of 4 or 5% of sucrose. The effect of different irradiance on the germination and early gametophyte development of D. sellowiana was analyzed in the autumn of 1998 (May until July). Essai de cryoconservation des gamétophytes de l'algue alimentaire Undaria pinnatifida (Laminariale) ArchiMer

Low temperature tolerance was determined for gametophytes of food alga Undaria pinnatifida according to various cryopreservation methods. Survival durations after storage in a medium containing 28% glycerol at −80 °C or in a 5-10% glycerol solution at −196 °C were only 9 and 4 days respectively. On the other hand, cooling at 5 °C/min. and storage at −15 °C for 15 minutes, without cryoprotective medium, did not reduce viability but this was highly decreased by storage for 15 minutes at −30 or −196 °C, with or without DMSO. The morphological cooling injuries were depigmentation and/or a contraction of the cells.

Gametophyte development and conservation of Ctenitis submarginalis (Dryopteridaceae) in Buenos Aires Province, Argentina Boletín de la Sociedad

The spore germination and gametophyte development of Ctenitis submarginalis from Punta Lara Natural Reserve, Buenos Aires province were studied as a contribution to the conservation of the species in this protected area. Spores collected from natural populations were sowing in Dyer medium and germinated in six day with a percentaje of 88 % fifteen days after sowing. The germination is Vittaria type and the gametophyte development is Aspidium type. The gametophyte of Ctenitis submarginalis is cordate, with a deep notch and well developed and symmetric wings. The gametangia appear 50 days after sowing. The sporophyte emerges 190 days after sowing and two different hairs were observed in the first lamina. In vitro spore culture and reproductive aspects of the annual fern Anogramma chaerophylla (Pteridaceae) Boletín de la Sociedad

Anogramma chaerophylla belongs to a fern genus with annual sporophytes and potentially perennating gametophytes. In the studied area, Natural Reserve Punta Lara, the plants grow stressed mainly by pollution and the invasion of exotic species. As a part of a project on conservation of ferns inhabiting Buenos Aires province, the objectives of this work were to evaluate in Anogramma chaerophylla the conditions for in-vitro spore germination and to analyse the different stages of its reproductive cycle. Spores were sterilized in an aqueous solution 10 % of NaCIO during different times and then sown in Petri dishes with Murashige & Skoog medium, without the addition of sucrose. Morphology and development of leaf papillae in the Pilotrichaceae Acta Botanica

Sporophyte morphology has been traditionally used in taxonomic studies of pleurocarpous mosses. However, the gametophyte morphology of the Pilotrichaceae still holds interesting and under-investigated characters. Here, we conducted an in-depth investigation of the morphology and development of Pilotrichaceae leaf papillae and assessed their usefulness in inferring the evolutionary history of the family. Specimens were observed under scanning electron microscopy, and we identified seven morphological types of papillae, as well as two different pathways of development. Our results suggest that papilla morphology and development are phylogenetically informative in the Pilotrichaceae.
